re-reading your post, I see you state that nothing was changed.  without access to the code, I don't know how to assist since this is not a known issue I am aware of.
 
You will need to submit a support request or engage a developer to go dig in and figure out the mystery.

When user is at checkout and logging in to their account using onepagecheckout.asp page, after entering their email address and password, an error comes up "Enter both your E-Mail and your Password." I have tested using my own email and password and same thing occurs. If we checkout using the Checkout.asp page, login works fine.

As far as I know nothing has been edited on the OPC page code, only changes to the languages.asp to change the text for guest checkout.

Any suggestions what would be happening?
